extend the axial imaging depth of the mantis microscope by moving the sample in z #20

The 3D volumes that we can acquire on the mantis microscope are limited to:

  • 20-30 um in the light-sheet arm. This limitation is set by the propagation depth of the light sheet for a given NA of illumination
  • ~60 um in the label-free arm. This limitation is set by the working range of the remote refocus optics and the travel range the scan mirror (which was picked to match the working range of the optics).

The overall axial imaging depth can be improved by moving the sample in Z using the ASI sample stage piezo and acquiring sets of 3D volumes with the remote refocus arms, which will later be stitched together. This will be important for imaging thicker samples such as zebrafish. Note that neuromasts fit within the remote refocus volumes, so this development is not urgent for now.
